5-HTTLPR is a polymorphism in the serotonin transporter gene.

There is a polymorphism rs25531 within the polymorphic region.

According to one of the first publications about the polymorphism the "short variant of the polymorphism reduces the transcriptional efficiency of the 5-HTT gene promoter, resulting in decreased 5-HTT expression and 5-HT uptake in lymphoblasts".[1]

It has been the target of personality genetics studies where one of the first publications hypothesized that 5-HTTLPR would be associated with neuroticism, and they found that subjects carrying one or two s-alleles were associated with higher neutoticism score compared to l-allele carriers.[2]
